Background :
	Interleukin-21 （IL-21）, expressed by activated CD4（+） T cells, is the newest member of the common gamma-chain family of cytokines, which includes IL-2, IL-4, IL-7, IL-9, IL-13, and IL-15. Its private receptor, IL-21R, is expressed on T, B, and NK cells. Within the B cell lineage, IL-21 regulates IgG1 production and cooperates with IL-4 for the production of multiple Ab classes in vivo. IL-21R has been shown to activate the Janus kinase/signal transducers and activators of transcription signaling pathway upon ligand binding. Initial studies have demonstrated that IL-21 has pleiotropic effects on the proliferation, differentiation, and effector functions of B, T, natural killer, and dendritic cells.
	Description :
	Recombinant Human IL-21 produced in E.Coli is a single, non-glycosilated polypeptide chain containing 133 amino acids and having a total molecular mass of 15,463 Dalton. The Recombinant Human IL-21 is purified by proprietary chromatographic techniques.
	Physical Appearance:
	Sterile Filtered White lyophilized （freeze-dried） powder.
	Formulation:
	Lyophilized from a concentrated （1mg/ml） solution containing no additives.
	Solubility:
	It is recommended to reconstitute the lyophilized Interleukin-21 in sterile 18MΩ-cm H₂O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
	Stability:
	Lyophilized Recombinant Human Interleukin-21 although stable at room temperature for 3 weeks, should be stored desiccated below -18⁰C. Upon reconstitution rHuIL-21 should be stored at 4⁰C between 2-7 days and for future use below -18⁰C. For long term storage it is recommended to add a carrier protein （0.1% HSA or BSA）. Please avoid freeze-thaw cycles.
	Purity:
	Greater than 95.0% as determined by: （a） Analysis by RP-HPLC. （b） Anion-exchange FPLC. （c） Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
	Amino acid sequence:
	The sequence of the first five N-terminal amino acids was determined and was found to be Gln-Gly-Gln-Asp-Arg.
	Dimers and aggregates:
	Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
	Biological Activity:
	ProSpec’s Recombinant IL-21 is fully biologically active when compared to standard. The ED50 as determined by the dose-dependant stimulation of human peripheral blood mononuclear cells is < 5ng/ml.
	Endotoxin:
	Less than 0.1 ng/µg （IEU/µg） of Recombinant Human IL-21.
	Protein content:
	Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280 nm using the absorbency value of 0.54 as the extinction coefficient for a 0.1% （1mg/ml） solution. This value is calculated by the PC GENE computer analysis program of protein sequences （InliGenetics）. 2. Analysis by RP-HPLC, using a standard solution of IL-21 as a Reference Standard.
	Usage:
